From 01. July 2025: Rabbit Pyrogen Test abolished – Are you prepared?

The European Pharmacopoeia (Ph. Eur.) is officially removing the Rabbit Pyrogen Test (RPT). From 1 July 2025, this method may no longer be used in accordance with supplement 11.8. This makes it clear that switching to animal-free methods is not only ethically necessary, but also mandatory from a regulatory perspective.

The key question for biopharmaceutical manufacturers is: Which alternative fulfils the regulatory requirements and is user-friendly and fast at the same time?

Our solution: NAT-MAT®

NAT-MAT® combines the established Monocyte Activation Test (MAT) with modern digital PCR (dPCR) technology. The system measures the gene expression of IL-1β and TNF-α and thus allows animal-free detection of pyrogen contaminations in pharmaceuticals according to Ph. Eur. 2.6.30.

  • Using a stable cell line ensures reproducible outcomes and a reliable cell supply

  • Normalizing the pyrogen concentration to the cell number delivers highly precise results

  • Rapid cell stimulation (4 hours) saves time

  • The highly automatable workflow saves time and reduces hand-on time to a minimum

  • Dual-cytokine detection increases dynamic range and enhances assay robustness

On 1 July 2025, an important change for biopharmaceutical quality control will come into force. For us, this date is also an opportunity to provide a special boost for all those who want to future-proof their test systems.

Look forward to 1 July – it's worth taking a closer look at NAT-MAT® on this day.

QUICK TIP

No touching allowed

When opening media bottles: Only twist the cap halfway off and leave it on the bottle – never place it on the table. These are often contaminated and even a brief touch can ruin your sample.
